Il nostro gruppo è interessato a comprendere come l'ambiente o le stimolazioni esterne possono influenzare la connettività cerebrale, la plasticità e il comportamento attraverso la regolazione dell'espressione genica durante il normale sviluppo o in modelli di malattie dello sviluppo neurologico e nelle tossicodipendenze. Il mio gruppo ha un'esperienza riconosciuta nello studio delle alterazioni neurologiche causate da mutazioni genetiche rare utilizzando modelli preclinici.

Sono più di quindici anni che studio le alterazioni molecolari, sinaptiche e comportamentali in modelli murini dei disturbi del neurosviluppo e dello spettro autistico, tra cui il deficit di CDKL5 e la sindrome di Rett, condizioni genetiche rare molto gravi.

Recentemente, abbiamo puntato la nostra attenzione sulla caratterizzazione di un nuovo modello murino del deficit di CDKL5, una malattia correlata alla sindrome di Rett ancora senza una cura, con l'obiettivo a lungo termine metterci al servizio questa comunità, pazienti e famiglie incluse, per di trovare approcci terapeutici.

Fin dal mio lavoro universitario, sono stato interessato a capire come l'ambiente e gli stimoli esterni possano influenzare la connettività e la plasticità del cervello, ed il comportamento, utilizzando approcci morfologici, fisiologici e molecolari. Il mio gruppo ha un'esperienza riconosciuta nella descrizione di alterazioni prodotte da mutazioni genetiche alla base delle funzioni sinaptiche e di gravi disturbi cerebrali.

Nel mio laboratorio, combiniamo abitualmente tecniche microscopiche ad alta risoluzione (per esempio microscopia confocale ed elettronica) con approcci molecolari, elettrofisiologici, biochimici e comportamentali per definire i processi meccanicistici alla base delle disfunzioni cerebrali. Abbiamo recentemente ampliato la nostra gamma di approcci sperimentali per includere: studi in vivo di popolazioni neuronali selettive nella corteccia cerebrale mediante microscopia a due fotoni, tracciamento virale di connessioni sinaptiche a lungo raggio, array di elettrodi multipli e registrazioni patch-clamp su neuroni primari coltivati e terapia genica.
